Karyotype Analysis

Human pluripotent stem cell cultures are prone to karyological changes. A major challenge of hPSC culture is maintaining the cells in the undifferentiated state while preserving a diploid karyotype. A normal diploid karyotype is one of the most important requirements for a truly pluripotent and untransformed ESC (Longo et al., 1997). Owing to the non-ideal culture systems for hESC lines, selection pressure is present in favor of chromosome duplications that confer an adaptive benefit (Draper et al., 2004; Andrews et al., 2005). For hESCs there appear to be common patterns of chromosome alteration representing “adaptation” of these cells to in vitro culture conditions, notably changes involving chromosomes 12 and 17 (Draper et al., 2004). For these reasons, we routinely karyotype our hESC lines every 5-8 passages to guard against the outgrowth of abnormal cells (Fig. 1). Karyotyping is a very labor-intensive assay. Much effort and skill is necessary to obtain good spreads and banding patterns that can be readily counted and analyzed. Karyotype by G-band and SKY spectral analysis

Our Karyotype Service

We provide Karyotype analysis of human stem cell lines by G-Band analysis. This analysis identifies chromosomal aberrations such as trisomies, deletions, translocations and inversions.
